本发明专利技术涉及一种生物酶法生产蚕蛹油并提取α-亚麻酸的方法,包括下列步骤:(1)烘干粉碎:蚕蛹经烘干粉碎过筛得蚕蛹粉;(2)水相酶解:按照料水质量比1:4~7将蚕蛹粉与水混合均匀,然后分两步进行酶解:首先,按照蚕蛹粉质量1.0~2.0%的比例加入胃蛋白酶,于30~40℃酶解2~4h,酶解过程中控制pH值为1.0~2.0,然后按照蚕蛹粉质量1.0~2.0%的比例加入木瓜蛋白酶,于55~65℃酶解2~4h,酶解过程中控制pH为6.0~7.0,酶解完成后酶解液加热至80~85℃灭酶;(3)离心、油水分离得到蚕蛹精油;(4)蚕蛹油脂肪酸制备;(5)尿素包埋:获得纯度70~75%的α-亚麻酸。本方法率先将生物酶法及尿素包埋等方法应用于蚕蛹细胞上,具有工艺简单、安全环保、成本低廉、所得蚕蛹油和α-亚麻酸质量优的优点。

生物酶法生产蚕蛹油并提取α?亚麻酸的方法,其特征在于:所述方法包括下列步骤:(1)烘干粉碎:蚕蛹经烘干粉碎过筛后,得到蚕蛹粉备用;(2)水相酶解:按照料水质量比1:4~7将蚕蛹粉与水混合均匀,然后分两步进行酶解:首先,按照蚕蛹粉质量1.0~2.0%的比例加入胃蛋白酶,于30~40℃酶解2~4h,酶解过程中控制pH值为1.0~2.0,然后按照蚕蛹粉质量1.0~2.0%的比例加入木瓜蛋白酶,于55~65℃酶解2~4h,酶解过程中控制pH为6.0~7.0,酶解完成后酶解液加热至80~85℃灭酶;(3)离心、油水分离:将酶解液离心分离得到残渣和上清液,上清液加热至50~60℃并静置进行物理破乳,然后将上清液进一步通过离心分离得到蚕蛹毛油和蚕蛹多肽液;(4)蚕蛹毛油精制:向蚕蛹毛油中加入2~4倍蚕蛹毛油体积的稀碱溶液,于30~40℃条件下搅拌1~3h,静置一段时间,待油水分离后,去除稀碱液,得到蚕蛹精油;(5)蚕蛹油脂肪酸制备:蚕蛹精油与1~1.2mol/L的氢氧化钠溶液按体积比1:3~5混合,于60~70℃小搅拌反应2~4h,静置冷却至常温,加水至澄清透明,调pH至2~3,加入蚕蛹精油3倍体积的石油醚或乙醚萃取,静置后收集醚层,水洗至中性,醚层经脱水后于40~50℃回收石油醚或乙醚,获得蚕蛹脂肪酸;(6)尿素包埋:将尿素与甲醇以体积比1:2~3混合,在60~70℃的条件下搅拌至充分溶解,加入适量的蚕蛹脂肪酸,尿素:甲醇:蚕蛹脂肪酸的体积比为1:2~3:0.6~1.0,搅拌2~3h混匀,然后冷却至室温,在?15~?30℃条件下静置包合20~35h,然后迅速减压抽滤,滤液调pH至2.0~3.0,用3~5倍滤液体积的石油醚或乙醚萃取,取醚层水洗至中性,经脱水后于40~50℃回收石油醚或乙醚,得到纯度70~75%的α?亚麻酸。...
