The chimeric antigen receptor coding gene is inserted into AAVS1 site of the iPS cells, and the resulting CAR iPS cells are differentiated into iCAR NK cells. The iCAR NK cells prepared by the invention are differentiated from the CAR iPS cells, which have long proliferative cycle and are more convenient for gene modification. Compared with iCAR NK cells acquired by lentiviral infection, iCAR NK obtained by differentiation of CAR iPS has a higher purity, which can be close to 100%. The gene insertion sites are AAVS1 loci, which have clear insertion location and higher safety. Because the iPS can proliferate almost infinitely, iCAR NK obtained by CAR iPS can also be obtained almost infinitely, and is not affected by peripheral blood isolation.
