The invention discloses a general formula compound with antineoplastic activity and a preparation method thereof, and also discloses the application of the compounds as antitumor agents. The general formula I, AA from valine, aspartic acid - 4 - cyclohexyl t-Boc, glutamine, lysine, leucine, alanine, tryptophan, phenylalanine, serine, O - benzyl isoleucine, glycine, histidine, methionine t-Boc, Gu An - 5 - cyclohexyl acid, asparagine, nitro arginine, proline, threonine or tyrosine residues. The present invention uses cell model to evaluate the antitumor activity of the compounds of formula I. The results show that the compounds of formula I. have excellent antitumor activity. Using S: 180 mice model to evaluate the antitumor effect of compounds of the general formula, the experimental results show that the compounds of formula I have excellent antitumor effect, can be clinically used as antitumor agent.
