一种天然骨胶原的制备方法,其特征在于,采用低温处理工艺,其包括如下步骤: 1)清洗:将新鲜畜禽骨进行水清洗; 2)粉碎:将清洗后的畜禽骨进行粉碎至粒度80~120目的骨泥; 3)脱脂、脱无机物:将骨泥与NaOH溶液或NaH CO3溶液进行混合,搅拌后分层,取底层,并用水进行洗涤,分离得蛋白质,再将其用石油醚或者丙酮浸提,浸提分离后的蛋白质中加入EDTA,搅拌均匀后,静置,然后用蒸馏水反复洗涤、过滤,得过滤物;再用盐酸胍-Tris-HCl混合搅拌,静置,然后用蒸馏水反复洗涤、过滤,得过滤物; 4)酸浸泡:将过滤物用酸浸泡; 5)酶水解:再加入蛋白酶进行酶解; 6)分离、纯化:然后离心分离,取上清液进行盐析,盐析的沉淀物用乙酸溶液溶解,再经盐析、膜透析处理得透析物; 7)冷冻 干燥:最后将透析物离心处理,并经冷冻干燥而成。
