The invention discloses a kind of targeted preparation of podophyllotoxin and its derivatives and a preparation method thereof, which comprises podophyllotoxin and its derivatives and acid-sensitive open-loop cucurbituril, in which the molar ratio of podophyllotoxin and its derivatives to acid-sensitive open-loop cucurbituril is 3-10:1. The open-loop cucurbituril has a C-type cavity and can form inclusion interaction with podophyllotoxin and its derivatives. Open-loop cucurbituril is a flexible molecule which can change the size of the cavity according to the size of the drug and form a good cavity matching with the drug, thus greatly enhancing the water solubility of the drug and facilitating the formation of the drug liquid preparations; moreover, the open-loop cucurbituril has pH stimulation responsiveness, and can release the drug autonomously under acidic conditions, thus achieving the targeted release of anti-cancer drugs. The development of new antineoplastic agents has potential application value.
