The invention discloses lhwox5 gene, which is a key gene for growth of main root of hybrid Liriodendron tulip, and its expression protein and application. The nucleotide sequence of lhwox5 gene, which is the key gene of the main root growth of Liriodendron hybrid, is shown in SEQ ID No.1, and the amino acid sequence of the expressed protein is shown in SEQ ID No.2. In this application, the expression of lhwox5 gene in different tissue parts of hybrid Liriodendron chinense was quantitatively analyzed by fluorescence quantitative PCR. The results showed that lhwox5 had high expression in stem tip, bud, root and other active parts. After lhwox5 gene is used to construct the vector, the lhwox5 gene is overexpressed in hybrid Liriodendron chinense by Agrobacterium transformation method. The results show that the main root of plants overexpressing lhwox5 gene is significantly shorter in hybrid Liriodendron chinense. It can be seen that lhwox5 gene has an important use in the growth of main root of hybrid Liriodendron chinense chinense.
