The invention relates to a method for improving the detection sensitivity of cocaine, which is characterized in that the nucleic acid aptamer for cocaine detection is broken into three sections, wherein the method comprises the following steps: (1) and three of cocaine aptamer probes to be mixed, the determination of cocaine form complexes with aptamer probe; (2) fluorescence detection, electrochemical detection and color detection. Since the length of the three nucleic acid probe is significantly lower than that of a one or two segment nucleic acid probe, the interaction between the probe and the other components of the probe and the detection system can cause background noise to be greatly reduced. Thus, the detection sensitivity of cocaine was improved significantly. Taking the detection of gold nanoparticles in cocaine as an example, the sensitivity of cocaine color detection was increased by an order of magnitude by using three nucleic acid probes. The method can be extended to the detection of various kinds of substances, including small molecules and heavy metal ions.
