The invention discloses a method for standardized multi-internal parameters combined quantification of exosome microRNAs and its application, which includes the following steps: (1) standardized processing of high-throughput microRNAs sequencing data; (2) screening internal parameters microRNAs by using coefficient of variation and standard deviation; (3) design of sequencing primers for digital PCR; (4) detection of microRNAs by digital PCR; (5) calculation of R_score value of multi-internal parameters combined standard. A large number of samples from healthy physical examination and patients with various degrees of disease were studied. It was found that microRNA 128_3p, microRNA 129_5p, microRNA 320A and Let_7i 5p or their combinations could be used as internal parameters for the standardization of microRNA in plasma exosomes with high stability and accuracy. Based on the digital PCR method, the invention provides an effective and convenient quantitative standardized detection for the study of microRNA in the exosome and in the exosome, makes up for the defect that there is no standard internal reference U6 snRNA in the exosome at present, and has important significance for thorough screening of the biomarkers of microRNA in the exosome.
