The invention belongs to the field of biotechnology. The present invention provides a recombinant protein, which contains all antigenic epitopes of gastrin 17 (hereinafter referred to as G 17), and repeats all antigenic epitope sequences through flexible fragments (four consecutive glycines) ten times to form the amino acid sequence of the recombinant G 17 protein. In order to increase the yield of the recombinant protein in prokaryotic expression system, the amino acid sequence of the recombinant protein was transformed into the corresponding nucleotide sequence using E. coli preferred codon. The nucleotide sequence was synthesized by chemical synthesis and the recombinant protein expression vector was constructed. The invention also relates to the preparation of the monoclonal antibody to the recombinant protein, and the monoclonal cell line is obtained after antigen immunization, cell fusion and multiple rounds of screening. The purified monoclonal antibodies were screened by protein-molecule interaction instrument, and labeled with fluorescent microspheres and coated with NC membranes respectively. The optimum combination of monoclonal antibodies was determined by experiments, which can be used for quantitative detection of G_17.
