柠檬酸生产麸制作过程中，柠檬酸发酵采用黑曲霉作为生产菌株，从菌种培养到发酵生产的过程可分为一级斜面培养、二级三角瓶麸曲培养、三级种子罐培养、发酵罐发酵。麸曲瓶一般是1000mL(或者2000mL)的三角玻璃瓶。三角瓶麸曲培养工艺过程非常复杂。取新鲜麸皮，用60目筛子筛去细粉，按麸皮:水＝1:1.0～1.3的比例加入水，拌匀至无干粉又无结团现象。拌匀后分装入1000mL的三角瓶中，每只1000mL的三角瓶装入麸皮40克(或者2000mL三角瓶，每只三角瓶装入100克麸皮)，用水洗麸皮表面，挤去水分，至用手抓有水而不下滴为宜，按适当量装瓶。用8层纱布封扎瓶口，在0.1MPa下灭菌40～60分钟，趁热摇散，冷却至35℃，培养一天，未发现气味异常或染菌，即可使用。在无菌室中，于无菌操作条件下，每一个三角瓶中接入一至二环己活化好的斜面菌种孢子，或2mL斜面菌种的孢子悬浮液，于30至32℃条件下培养16～20小时后，白色小菌落己盖满曲层表面，这时应摇瓶翻曲一次，使培养基疏松、铺平、继续培养。再经4至6小时，即接种24...
