The embodiment of the invention discloses a method for extracting and detecting polyphenols from `Fengdan'peony based on response surface optimization, which includes: taking the young leaves of `Fengdan' peony as raw materials, vacuum drying and crushing to obtain treated powder; centrifuging the treated powder after enzymatic hydrolysis under different technological conditions, retaining supernatant, centrifuging the sediment after water extraction, obtaining supernatant and mixing it twice. Polyphenol extract was obtained from clear liquid and constant volume; single factor variable test was carried out with enzyme concentration, enzymatic hydrolysis temperature, enzymatic hydrolysis time and pH as the factors to investigate. Response surface design was carried out with polyphenol extraction rate (Y) as the response value, and the optimal extraction process was obtained. The main chemical components of polyphenol extract from leaves were analyzed and determined by HPLC. The invention is a response surface optimization process for extracting polyphenols from leaves of `Fengdan'peony with high extraction efficiency, accurate technological conditions and simple operation. An HPLC method for detecting polyphenols in leaves of `Fengdan' peony is established, and the main chemical composition and content of polyphenols are determined.
