The invention discloses the application of short-leaf hematoxylin acid methyl ester in the preparation of anti-influenza virus drugs. The inventor found that methyl hematoxylate had a strong anti-influenza A virus H1N1 subtype effect in the range of 0.78 100 ug/mL. The IC50 value of methyl hematoxylate was 18.90 5.65 micromol/L, which was slightly lower than that of ribavirin (IC50 = 0.56 1.10 micromol/L). Plaque assay showed that methyl hematoxylate inhibited the plaque formation of MDCK cells (canine renal epithelial cells) induced by H1N1 virus in a concentration-dependent manner. Methyl hematoxylate can inhibit the expression of viral NP protein and PB2 polymerase in MDCK cells, and the inhibition increases with the increase of concentration. Meanwhile, methyl hematoxylate also inhibited H3N2, with IC50=71.74+1.48 ug/ml.
