The invention discloses a mannanase mutant with improved heat resistance and its gene and application. The GH5 mannanase PMan5A family derived from Penicillium sp. WN1 is used as female parent, and histidine (H), phenylalanine (F), leucine (L) and leucine (L) at sites 93, 94, 356 and 389 of the female parent are used as molecular biological means. Alanine (A) mutated into tyrosine (Y), tyrosine (Y), histidine (H) and proline (P). The results showed that the heat tolerance of single-point mutants H93Y, L356H and A389P was significantly higher than that of wild-type enzymes PMan5A, and the heat tolerance of combination mutants H93Y/F94Y, H93Y/L356H, H93Y/A389P, L356H/A389P, H93Y/F94Y/L356H, H93Y/L356H/A389P, H93Y/F94Y/L356H/A389P and H93Y/F94Y/L356H/A389P showed the superposition effect of single-point mutation. Loci 93, 94, 356 and 389 of 5A play an important role in the thermostability of GH5 mannanases, which may be of great significance to the study of thermostability mechanism of other GH5 (alpha/beta) 8 barrel enzymes.
