The invention discloses a reagent for detecting YAP K342 methylation and its application. YAP K342 methylation modification is specifically disclosed as a marker for tumor diagnosis and prognosis analysis, especially for the diagnosis and prognosis of lung and bowel cancer tumors. It was found that the monomethylation site K342 of YAP occurred and the methylation modified polyclonal antibody K342 was prepared. The experimental results showed that the methylation of YAP K342 showed specific high expression in lung cancer and intestinal cancer, but there was no such specific high expression in normal tissues. Moreover, the K342 methylation modification was negatively correlated with the prognosis of tumor patients. Therefore, YAP K342 methylation can be used as a marker of tumor diagnosis and prognosis analysis, especially as a marker of lung cancer and intestinal cancer diagnosis, can clearly characterize the occurrence of tumors and poor prognosis. The invention provides an important reference basis for clinical diagnosis and treatment.
