A method based on ITS2 for identifying Panax japonicus and its non-homologous adulterants discloses a method for identifying Panax japonicus and its non-homologous adulterants at DNA level. The method comprises the following steps: 1) extracting the DNA of Panax japonicus from the sample to be tested; 2) amplifying the fragments of ITS2 sequence with the DNA of the sample to be tested; 3) positively sequencing the PCR products, and then sequencing them. ITS2 sequences were obtained by ITS2 database cutting; 4) NJ phylogenetic trees were constructed, average genetic distances within and between species were analyzed, and secondary structural characteristics were compared. The results showed that ITS2 sequences were clustered together, and the average genetic distances within species of Panax japonicus were significantly smaller than those among the hybrids with different genus, and the secondary structure of Panax japonicus was significantly different from that of the hybrids with different genus. This indicated that ITS2 sequences could be used to identify Panax japonicus and its hybrids quickly and accurately. . The method has the advantages of simple operation, high resolution, strong applicability and repeatability, and can quickly and accurately identify Panax japonicus and its adulterants.
